Onyx’s commercial portfolio includes an established therapy approved for both advanced kidney cancer and liver cancer, where it remains the only approved targeted systemic therapy. Building upon this portfolio, Onyx, with its collaborator Bayer, is actively pursuing new indications for this molecule, including lung cancer, thyroid cancer, breast cancer, colorectal cancer, ovarian cancer, and as an adjuvant therapy for liver cancer and kidney cancer.
Onyx is also actively engaged in a proteasome inhibition development program. The proteasome has been validated as an important clinical target in cancer, and Onyx is developing next-generation proteasome inhibitors with a high degree of selectivity, with the goal of increasing therapeutic efficacy and reducing off-target toxicities. Based on results from the Phase 2b 003-A1 study evaluating our lead product candidate in this program in patients with relapsed and refractory multiple myeloma, Onyx has initiated a rolling submission of a New Drug Application (NDA) for potential accelerated approval in the U.S. and initiated two Phase 3 trials to support full registration in the U.S. and Europe.
The Onyx development program also includes an oral proteasome inhibitor that is currently in Phase 1 testing in solid tumors; an immunoproteasome inhibitor with activity in preclinical models of autoimmune disorders; an alpha-folate receptor targeted inhibitor of thymidylate synthase that is in Phase 1 testing; and an oral small molecule cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, now in Phase 2 clinical development, resulting from a collaboration with Warner-Lambert Company, now Pfizer.
